SPRi, or surface plasmon resonance microscopy (SPRM), is a label-free, surface sensitive spectroscopic technique. It is used to detect the binding of biological molecules onto arrays of probe biomolecules covalently attached to chemically-modified gold surfaces. Or more specifically, SPRi has been developed to examine affinity between biomoleculars, screen biomarkers and detect biopsy specimens.
Surface Plasmon has already been applied in a number of studies ranging from gas sensing to biosensing, from immuno-sensing to electrochemical studies. As to how SPRi works, “a plasmon can be thought of as a ray of light bound onto a surface – propagating among the surface and presenting itself as an electromagnetic field,” explained the leading Scientist of Creative Peptides’ SPRi team.
“Surface Plasmon Resonance (SPR) is actually an optical detection process that occurs when a polarized light hits a prism covered by a thin planar metal (typically gold or silver) layer. By employing this technique, the association and dissociation of biomolecules can be monitored,” the Scientist further added.
With the above-mentioned characteristics and advantages, SPR has been chosen by drug researchers as a method to select and characterize therapeutics, while for those engaged in basic life sciences, SPR is used as means to investigate certain events like biological binding. When combined with other disciplines such as surface chemistry and software, there is still more to discover as to what SPR could do.
Overally speaking, Surface Plasmon Resonance imaging is already well-established as a leading technology in measuring binding association (ka) and dissociation rates (kd), affinities (KD), such as peptide/antibody, antibody/bacteria.
